I would like to be able to toggle the preview mode of the glyph window on and off rather than having to hold the ~ key to keep it on. This would be very useful when I just want to look over a font with complex overlaps, masks, etc. letter-by-letter without having to hold the preview button down the entire time.

I don't think there's a way to do this in the Glyph window, but if you open the glyph in the Metrics window, enlarge it to a sufficient size, click on it, and walk through your glyphs using the keyboard short cut (command right- and left-bracket on Mac), you can get a similar effect. When you get to a glyph you want to edit, just double click.
